Educational Mobility, Pace of Aging, and Lifespan Among Participants in the Framingham Heart Study

From the abstract: " Is upward educational mobility associated with a slower pace of biological aging and increased longevity? In this cohort study of 3101 participants representing 2 generations of the Framingham Heart Study, upward educational mobility was associated with a slower pace of aging (as measured with whole-blood DNA-methylation data) and lower risk of death. Slower pace of aging accounted for approximately half of the association between educational mobility and mortality. These results suggest that interventions to promote educational attainment may slow the pace of biological aging and promote longevity. "

Such biomarkers could predict aging-related outcomes and could serve as surrogate endpoints for the evaluation of interventions promoting healthy aging and longevity. However, no consensus exists on how biomarkers of aging should be validated before their translation to the clinic. Here, we review current efforts to evaluate the predictive validity of omic biomarkers of aging in population studies, discuss challenges in comparability and generalizability. "


Mendelian randomization evidence for the causal effects of socio-economic inequality on human longevity among Europeans.

Here we performed two-sample Mendelian randomization analyses applying genetic instruments of education, income and occupation (n?=?248,847 to 1,131,881) to estimate their causal effects and consequences on parental lifespan and self-longevity (n?=?28,967 to 1,012,240) from the largest available genome-wide association studies in populations of European ancestry. Each 4.20 years of additional educational attainment were causally associated with a 3.23-year-longer parental lifespan independently of income and occupation and were causally associated with 30–59% higher odds of self-longevity.


Epigenetic profile of Japanese supercentenarians: a cross-sectional study

Epigenome-wide association analyses showed that centenarians and supercentenarians had younger-than-expected epigenetic states (DNA methylation profiles similar to those of non-centenarians) for 557 CpG sites enriched in cancer-related and neuropsychiatric-related genes, whereas these individuals had advanced (or older) epigenetic states for 163 CpG sites represented by genes related to TGF-ß signalling, which is involved in anti-inflammatory responses and known to contribute to healthy ageing.


Diet modulates genetic effects on longevity.

Why some individuals live longer than others is of major interest across a range of scientific disciplines, from evolutionary biology to human health. Evolutionary theory tells us that lifespan-reducing alleles should be purged from the gene pool1, yet decades of genome-wide association studies (GWAS) and model organism studies have shown that such alleles persist. Here, we address one potential explanation for this conundrum, the idea that alleles that regulate lifespan do so only in certain contexts (known as ‘genotype-by-environment’ (GxE) effects).


Analysis of Epigenetic Age Acceleration and Healthy Longevity Among Older US Women

This cohort study was a secondary analysis of 3 Women’s Health Initiative ancillary studies among 1813 women eligible to survive to age 90 years by end of study period. The study found that increased epigenetic age acceleration as measured by 4 epigenetic clocks was associated with lower odds of survival to age 90 years with intact mobility; results were similar when including intact cognitive functioning. Meaning These findings suggest that epigenetic age acceleration may be a useful biomarker to estimate functional and cognitive aging among older women.

We used genome-wide association study (GWAS) summary statistics for ADHD (n?=?53?293) and 124 traits related to anthropometry, cognitive function and intelligence, early life exposures, education and employment, lifestyle and environment, longevity, neurological, and psychiatric and mental health or personality and psychosocial factors available in the MR-Base database (16?067 =n?=766?345). Our findings strengthen previous evidence of a causal effect of ADHD liability on smoking and major depression, and are consistent with a causal effect on odds of decreased average total household income [odds ratio (OR)?=?0.966, 95% credible interval (CrI)?=?(0.954, 0.979)] and increased lifetime number of sexual partners [OR?=?1.023, 95% CrI?=?(1.013, 1.033)].


Making sense of the ageing methylome.

Over time, the human DNA methylation landscape accrues substantial damage, which has been associated with a broad range of age-related diseases, including cardiovascular disease and cancer. Various age-related DNA methylation changes have been described, including at the level of individual CpGs, such as differential and variable methylation, and at the level of the whole methylome, including entropy and correlation networks. Here, we review these changes in the ageing methylome as well as the statistical tools that can be used to quantify them. We detail the evidence linking DNA methylation to ageing phenotypes and the longevity strategies aimed at altering both DNA methylation patterns and machinery to extend healthspan and lifespan. Lastly, we discuss theories on the mechanistic causes of epigenetic ageing.
